Description
Overview of the medicine
Esomeprazole is a proton pump inhibitor that reduces acid secretion in the stomach. It is used to treat conditions like gastroesophageal reflux disease (GERD), ulcers, and Zollinger-Ellison syndrome.
Uses & Indications
Dosage
Adults
Erosive Esophagitis: 20 or 40 mg once daily for 4-8 weeks. GERD (maintenance): 20 mg once daily. H. pylori eradication: 40 mg once daily for 10 days (with amoxicillin and clarithromycin). Zollinger-Ellison syndrome: Initial 40 mg twice daily, adjusted as needed.
Elderly
No dosage adjustment generally needed, but use with caution due to potential age-related physiological changes.
Renal_impairment
No dosage adjustment needed for mild to moderate renal impairment. For severe impairment, consult a doctor.
How to Take
Take orally, preferably one hour before a meal. Swallow the whole tablet; do not chew, crush, or break it.
Mechanism of Action
It irreversibly blocks the H+/K+-ATPase (proton pump) in gastric parietal cells, thereby inhibiting the final step of acid production.
Pharmacokinetics
Onset
Within 1 hour
Excretion
Primarily via urine (approx. 80%) and feces (approx. 20%).
Half life
Approximately 1-1.5 hours
Absorption
Rapidly absorbed after oral administration, bioavailability is about 50-68%. Food may delay absorption but does not significantly affect bioavailability.
Metabolism
Extensively metabolized in the liver by the CYP2C19 and CYP3A4 enzyme systems.
