Description
Overview of the medicine
Gatifloxacin 0.3% eye drop is an ophthalmic antibiotic used to treat bacterial conjunctivitis caused by susceptible strains of microorganisms. It belongs to the fluoroquinolone class.
Uses & Indications
Dosage
Adults
Instill 1 drop into the affected eye(s) every 2 hours while awake, up to 8 times a day, for the first 2 days, then 1 drop 4 times a day for the next 5 days.
Elderly
No dosage adjustment necessary.
Renal_impairment
No dosage adjustment necessary as systemic absorption is minimal.
How to Take
For ophthalmic use only. Tilt head back, pull down the lower eyelid to form a pouch, instill the drop, and close the eye gently. Do not touch the dropper tip to any surface.
Mechanism of Action
Gatifloxacin, a fluoroquinolone, inhibits bacterial DNA gyrase and topoisomerase IV, essential enzymes involved in bacterial DNA replication, transcription, repair, and recombination. This leads to cell death.
Pharmacokinetics
Onset
Rapid onset of action, typically within hours for antibacterial effect.
Excretion
Primarily excreted unchanged in urine if absorbed systemically, but topical application leads to negligible systemic excretion.
Half life
Systemic half-life is around 7-8 hours, but topical application leads to very low systemic exposure. Ocular tissue concentrations are maintained for several hours.
Absorption
Minimal systemic absorption after topical ophthalmic administration. Peak plasma concentrations are generally below quantifiable limits.
Metabolism
Minimally metabolized systemically.
Side Effects
Contraindications
- Hypersensitivity to gatifloxacin, other quinolones, or any component of the medication
- Viral or fungal eye infections
Drug Interactions
Systemic medications
No significant systemic drug interactions expected due to minimal systemic absorption.
Other ophthalmic preparations
If using multiple eye drops, administer them at least 5 minutes apart.
Storage
Store at room temperature (20°C to 25°C), away from light and moisture. Do not freeze. Keep out of reach of children.
Overdose
Overdose with ophthalmic drops is unlikely due to the small amount of drug per drop and minimal systemic absorption. If topical overdose occurs, the eye can be rinsed with lukewarm water.
Pregnancy & Lactation
Pregnancy Category C. Use only if potential benefit outweighs risk. Caution advised during lactation; systemic absorption is minimal, but monitor infant for adverse effects.
