Description
Overview of the medicine
Krimizole-DS 400 mg Chewable Tablet is an anthelmintic medication used to treat a variety of parasitic worm infections in humans. It works by preventing the worms from absorbing sugar (glucose), which is essential for their survival, thereby killing them. This chewable form is often preferred for ease of administration, especially in children and individuals who have difficulty swallowing tablets.
Uses & Indications
Dosage
Adults
For common intestinal worms: 400 mg as a single dose. For specific infections like neurocysticercosis or hydatid disease, dosage and duration vary significantly and require medical supervision.
Elderly
No specific dose adjustment is generally required for elderly patients with normal renal and hepatic function. Use with caution in patients with hepatic impairment.
Renal_impairment
No dose adjustment is generally required for renal impairment, but caution is advised in patients with severe renal disease.
How to Take
This is a chewable tablet. It should be chewed thoroughly before swallowing. It can be taken with or without food, but taking it with a fatty meal may enhance absorption. For systemic infections, it is often recommended to be taken with food.
Mechanism of Action
Albendazole, a benzimidazole carbamate, exerts its anthelmintic effect by inhibiting tubulin polymerization, leading to the disruption of microtubule formation in the intestinal and tegumental cells of the worms. This impairs glucose uptake by the worms, depleting their glycogen stores and ultimately leading to their immobilization and death.
Pharmacokinetics
Onset
Variable, depending on the type of infection; typically within hours to days for anthelmintic effects.
Excretion
Mainly excreted in the urine as metabolites (less than 1% as unchanged albendazole sulfoxide); some biliary excretion.
Half life
Albendazole sulfoxide: approximately 8-12 hours.
Absorption
Poorly absorbed from the gastrointestinal tract (less than 5%). Absorption is increased with fatty meals. Rapidly converted to its active metabolite, albendazole sulfoxide.
Metabolism
Extensive first-pass metabolism in the liver to its primary active metabolite, albendazole sulfoxide, and then to other inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to Albendazole or other benzimidazoles
- Pregnancy (especially first trimester)
Drug Interactions
Ritonavir
May decrease albendazole sulfoxide levels.
Cimetidine
Increases plasma concentrations of albendazole sulfoxide.
Praziquantel
Increases plasma concentrations of albendazole sulfoxide.
Dexamethasone
Increases plasma concentrations of albendazole sulfoxide.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptomatic and supportive treatment should be initiated. There is no specific antidote for albendazole overdose. Gastric lavage and activated charcoal may be considered if ingestion is recent.
Pregnancy & Lactation
Albendazole is generally contraindicated in the first trimester of pregnancy due to potential teratogenicity. In later trimesters, it should only be used if the potential benefit justifies the potential risk to the fetus. It is unknown whether albendazole or its metabolites are excreted in human milk, so caution should be exercised during lactation.
