Description
Overview of the medicine
Debolin 2000 IU Tablet is a vitamin D3 supplement used to treat and prevent vitamin D deficiency, promote calcium absorption, and support bone health.
Uses & Indications
Dosage
Adults
Typical maintenance dose is 2000 IU daily. For deficiency, higher doses (e.g., 4000 IU daily or weekly/monthly regimens) may be prescribed by a physician based on serum 25(OH)D levels.
Elderly
Similar to adults, but may require higher doses due to reduced skin synthesis and absorption. Individual assessment recommended.
Renal_impairment
Monitor vitamin D and calcium levels. May require activated forms of Vitamin D (e.g., calcitriol) under medical supervision, as renal conversion of cholecalciferol to calcitriol may be impaired.
How to Take
Administer orally, preferably with a meal to enhance absorption, particularly if the meal contains fat.
Mechanism of Action
Cholecalciferol is converted to its active form, calcitriol, which regulates calcium and phosphate homeostasis. It promotes absorption of calcium and phosphate from the small intestine, reabsorption of calcium in the kidneys, and plays a crucial role in bone mineralization.
Pharmacokinetics
Onset
Significant increase in serum 25(OH)D levels typically observed within days to weeks; full clinical effects may take longer.
Excretion
Primarily excreted via bile into feces; a small amount is excreted in the urine.
Half life
Cholecalciferol (unmetabolized) has a half-life of 1-2 days. The active metabolite, 25(OH)D (calcifediol), has a half-life of 2-3 weeks.
Absorption
Well absorbed from the small intestine, primarily in the presence of bile salts and dietary fat.
Metabolism
Hydroxylated in the liver to 25-hydroxyvitamin D3 (calcifediol), then further hydroxylated in the kidneys to 1,25-dihydroxyvitamin D3 (calcitriol), the active form.
Side Effects
Contraindications
- Hypercalcemia (high calcium levels in blood)
- Vitamin D toxicity
- Hypersensitivity to cholecalciferol or any excipients of the tablet
- Nephrolithiasis (kidney stones) or nephrocalcinosis (calcium deposition in kidneys)
Drug Interactions
Digoxin
Hypercalcemia caused by vitamin D may exacerbate the toxic effects of digoxin.
Corticosteroids
May reduce the effects of vitamin D by decreasing calcium absorption and increasing vitamin D metabolism.
Thiazide diuretics
May reduce urinary calcium excretion, increasing the risk of hypercalcemia.
Anticonvulsants (e.g., phenytoin, barbiturates)
May increase the metabolism of vitamin D, potentially reducing its effectiveness.
Cholestyramine, Colestipol, Orlistat, Mineral oil
May impair absorption of vitamin D.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose (hypervitaminosis D) are primarily due to hypercalcemia and may include nausea, vomiting, constipation, anorexia, polyuria, polydipsia, weakness, headache, and confusion. Severe cases can lead to kidney damage, soft tissue calcification, and cardiac arrhythmias. Management involves discontinuing vitamin D, hydration, low calcium diet, and potentially corticosteroids or bisphosphonates.
Pregnancy & Lactation
Generally considered safe at recommended daily allowances during pregnancy and lactation. Higher doses should only be used under strict medical supervision and if the potential benefit outweighs the risk. Vitamin D is essential for fetal bone development and is secreted into breast milk.

YesClinical Trials
Numerous clinical trials and studies have demonstrated the efficacy of cholecalciferol in treating and preventing vitamin D deficiency and its associated conditions. Specific trials for the 'Debolin' brand are typically bioequivalence studies.
Lab Monitoring
- Serum 25(OH)D levels (to assess vitamin D status)
- Serum calcium levels (to monitor for hypercalcemia)
- Serum phosphate levels
- Renal function tests (e.g., creatinine) with long-term high-dose therapy
Doctor Notes
- Assess baseline serum 25(OH)D levels before initiating high-dose vitamin D therapy.
- Monitor serum calcium, phosphate, and renal function, especially in patients receiving high doses, with pre-existing renal impairment, or on concomitant medications like thiazide diuretics.
- Advise patients on appropriate sun exposure and dietary sources of vitamin D to complement supplementation.
Patient Guidelines
- Take exactly as prescribed by your doctor or as indicated on the package.
- Do not exceed the recommended dose without medical advice.
- Inform your doctor about all other medications and supplements you are taking.
- Report any symptoms of hypercalcemia (e.g., severe nausea, vomiting, unusual tiredness) to your doctor immediately.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and resume your regular dosing schedule. Do not take a double dose to make up for a missed one.
Driving Precautions
Debolin 2000 IU Tablet is not known to affect the ability to drive or operate machinery. No special precautions are necessary.
Lifestyle Advice
- Maintain a balanced diet rich in calcium and vitamin D, if possible.
- Ensure adequate sun exposure, while protecting skin from excessive UV radiation.
- Regular exercise can contribute to bone health.
