Description
Overview of the medicine
Hybrox 75 mg Capsule contains Pregabalin, which is primarily used to treat neuropathic pain, fibromyalgia, and as an adjunct therapy for partial onset seizures in adults. It works by calming damaged or overactive nerves in the body.
Uses & Indications
Dosage
Adults
Starting dose usually 75 mg twice daily, may be increased up to 300 mg twice daily depending on response and tolerability. For some indications, up to 600 mg/day in divided doses.
Elderly
No specific dose adjustment needed based on age alone, but renal function should be assessed.
Renal_impairment
Dosage must be adjusted based on creatinine clearance. Lower doses are required for patients with impaired renal function.
How to Take
Take orally with or without food. Swallow the capsule whole with water. Do not crush, chew, or open the capsule.
Mechanism of Action
Pregabalin binds to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system, reducing the release of several neurotransmitters like glutamate, norepinephrine, and substance P. This action is believed to modulate the overactivity of neurons responsible for pain signaling and seizure generation.
Pharmacokinetics
Onset
Within 1 week for neuropathic pain; varies for other indications.
Excretion
Primarily excreted unchanged in the urine (approximately 98%).
Half life
Approximately 6.3 hours.
Absorption
Rapidly absorbed after oral administration, reaching peak plasma concentrations within 1.5 hours.
Metabolism
Negligible hepatic metabolism; does not induce or inhibit liver enzymes.
Side Effects
Contraindications
- Hypersensitivity to Pregabalin or any component of the formulation.
- Severe renal impairment without dose adjustment.
Drug Interactions
CNS Depressants (e.g., Opioids, Benzodiazepines, Alcohol)
May enhance the central nervous system (CNS) depressant effects of Pregabalin, leading to increased sedation, dizziness, and respiratory depression.
Thiazolidinedione Antidiabetic Agents (e.g., Rosiglitazone, Pioglitazone)
May increase the risk of weight gain and peripheral edema when co-administered with Pregabalin.
Storage
Store at room temperature (below 30°C) in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, agitation, and restlessness. Management is primarily supportive; gastric lavage may be considered soon after ingestion. Hemodialysis can remove Pregabalin from the plasma.
Pregnancy & Lactation
Pregnancy Category C. Should only be used if the potential benefit justifies the potential risk to the fetus. Pregabalin is excreted in breast milk; therefore, caution should be exercised when administered to a nursing woman.
