Description
Overview of the medicine
Liana 3 mg tablet contains Dienogest, a progestin, primarily used for the treatment of endometriosis and as an oral contraceptive. It helps in reducing pain associated with endometriosis and regulates the menstrual cycle.
Uses & Indications
Dosage
Adults
One 3 mg tablet daily without interruption, preferably at the same time each day, regardless of food intake.
Elderly
Not indicated for postmenopausal women. No specific dosage adjustment for elderly women prior to menopause is required, but clinical data are limited.
Renal_impairment
No specific dosage adjustment is recommended, but caution is advised in severe renal impairment.
How to Take
Take one tablet daily, preferably at the same time each day, with or without food. Swallow the tablet whole with a sufficient amount of liquid. Do not chew or crush.
Mechanism of Action
Dienogest is a 19-norprogestin derivative with high progestogenic activity and moderate antiandrogenic activity. It reduces the endogenous production of estradiol, thereby suppressing the trophic effects of estradiol on both eutopic and ectopic endometrium. It also has a local progestogenic effect on the endometrium, leading to decidualization and subsequent atrophy of endometrial lesions. For contraception, it suppresses ovulation.
Pharmacokinetics
Onset
Within 24 hours (for endometrial effects, symptomatic relief may take weeks)
Excretion
Mainly excreted via the kidneys (primarily as metabolites) and to a lesser extent via feces.
Half life
Approximately 9-10 hours
Absorption
Rapidly and almost completely absorbed after oral administration. Peak plasma concentrations are reached about 1.5 hours after intake.
Metabolism
Extensively metabolized by the cytochrome P450 3A4 system (CYP3A4) into pharmacologically inactive metabolites.

CYP3A4 Inducers (e.g., Rifampicin, Phenytoin, Carbamazepine)
May reduce Dienogest plasma concentrations and therapeutic effects.
CYP3A4 Inhibitors (e.g., Ketoconazole, Erythromycin, Grapefruit juice)
May increase Dienogest plasma concentrations, leading to increased side effects.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Overdose may cause nausea, vomiting, and slight vaginal bleeding. No specific antidote exists. Treatment should be symptomatic.
