Description
Overview of the medicine
Indomethacin is a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain, swelling, and fever. Lucidol 100 mg Suppository is a rectal formulation primarily used for conditions such as arthritis, gout, and other inflammatory disorders.
Uses & Indications
Dosage
Adults
Typically 50-100 mg rectally, 1-2 times daily. Max 200 mg/day. Dosage should be individualized and lowest effective dose used.
Elderly
Lower doses may be required due to increased risk of side effects and impaired renal function. Start with 50 mg once daily.
Renal_impairment
Reduced dosage and careful monitoring are necessary due to risk of renal toxicity. Avoid in severe renal impairment.
How to Take
For rectal use only. Remove the suppository from its wrapper and insert it into the rectum. Lie down for a few minutes after insertion.
Mechanism of Action
Inhibits cyclooxygenase (COX-1 and COX-2) enzymes, thereby reducing prostaglandin synthesis, which are mediators of pain, inflammation, and fever.
Pharmacokinetics
Onset
Within 30 minutes to 1 hour, full effect within a few hours.
Excretion
Excreted primarily in urine (60%) and feces (33%) as metabolites and some unchanged drug.
Half life
Approximately 4.5 hours, but can vary.
Absorption
Well absorbed from the rectum, though onset may be slower than oral forms. Peak plasma concentrations are reached within 2-4 hours.
Metabolism
Primarily in the liver by O-demethylation and N-deacylation, followed by glucuronidation.

Storage

Overdose
Symptoms may include nausea, vomiting, epigastric pain, headache, drowsiness, and central nervous system depression. Management is supportive and symptomatic, including gastric lavage or induced emesis if oral overdose, but for suppository, symptomatic treatment is key. Monitor vital signs and renal function.
