Description
Overview of the medicine
K-Lac 30 mg Injection is a non-steroidal anti-inflammatory drug (NSAID) used for the short-term management of moderately severe acute pain requiring opioid-level analgesia.
Uses & Indications
Dosage
Adults
Initial dose 30 mg IM or IV, followed by 30 mg every 6 hours, not exceeding 120 mg/day for IM or 60 mg/day for IV in patients >65 kg. Maximum duration of treatment is 5 days.
Elderly
Reduce dose to 15 mg every 6 hours (max 60 mg/day).
Renal_impairment
Contraindicated in advanced renal impairment. For mild to moderate, dose reduction to 15 mg every 6 hours (max 60 mg/day).
How to Take
Administer intravenously or intramuscularly. IV doses should be administered over a minimum of 15 seconds. IM doses should be given slowly and deeply into a large muscle.
Mechanism of Action
Ketorolac inhibits prostaglandin synthesis by reversibly inhibiting cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzymes. This results in reduced inflammation, pain, and fever.
Pharmacokinetics
Onset
Analgesic effect usually within 30 minutes after IM/IV administration.
Excretion
Primarily renal (approximately 91%), with some fecal excretion (6%).
Half life
Approximately 4-6 hours (varies with age and renal function).
Absorption
Rapid and complete absorption after IM administration. Peak plasma concentration achieved within 30-60 minutes.
Metabolism
Primarily hepatic via hydroxylation and glucuronidation.

Increased lithium plasma levels and toxicity.
Diuretics
Reduced natriuretic effect and increased risk of renal impairment.
Methotrexate
Increased methotrexate toxicity.
Other NSAIDs or aspirin
Increased risk of GI adverse effects.
Warfarin, Heparin, other anticoagulants
Increased risk of bleeding.
